Effet de la colchicine sur les mouvements des stomates et l'ultrastructure des cellules stomatiques de Tradescantia virginiana

Résumé

Dans les stomates de Tradescantia virginiana L. en cours d'ouverture, des microtubules sont souvent visibles ce qui n'est pas le cas dans les stomates ouverts ou fermés à l'équilibre. Ces microtubules, situés dans le cytoplasme pariétal, sont principalement observables sur les coupes paradermales tangentlelles, et se situent dans le prolongement des villosités du plasmalemme et de la paroi. La colchicine, employée à la concentration de 1 mM et 10 mM, inhibe l'ouverture des stomates placés en conditions d'ouverture. Cette drogue inhibe l'addition des molécules de tubuline, ce qui conduit à la dépolymérisation des microtubules. Les modifications ultrastructurales dues à la colchicine sont décrites. Ces résultats conduisent à penser que la présence et l'Intégrité des microtubules sont indispensables à l'ouverture des stomates.

Summary

In stomata of Tradescantia virginiana L., microtubules were observed in guard cells during stomatal opening, but never in open or closed stomata. These microtubules were present along the plasmalemma and in the parietal cytoplasm on the paradermal section. They were located near the wall foldings. Colchicine 1 mM and 10 mM inhibited the movement during stomatal opening. This drug Inhibits the addition of tubulin molecules to microtubules, leading to microtubule depolymerization. Ultrastructural variations of guard cells produced by colchicine were examined. These results suggest that microtubules might be involved in the stomatal opening.
