A loss-of-function of TDP-43 in the nucleus accelerates pathological TDP-43 release via autophagosome-derived vesicles in the pathological donor cell (left). EV containing pathological TDP-43 is taken up in the healthy recipient cell (right). Pathological TDP-43 promotes the convertion of newly translated or nuclear-exported endogenous TDP-43 into the pathological form, which cause the de novo TDP-43 pathology.
